Stem tip meristematic tissue culture and rapid propagation technology of Ipomoea batatas variety Xinxiang

Abstract:

To establish the meristematic tissue culture and rapid propagation system of Ipomoea batatas stem tip,the variety of Xinxiang was used as the material and its stem tip meristematic tissue was stripped to cultivate detoxified seedlings,and MS was used as the basal medium,with the addition of different mass concentrations of 6-BA and NAA,to screen the most suitable medium for bud induction and proliferation rooting of stem tip meristematic tissue culture by a two-factor randomized block test. The results showed that the suitable medium for bud induction of stem tip meristematic tissue of Ipomoea batatas variety Xinxiang was MS+0.5 mg·L-1 6-BA +0.1 mg·L-1 NAA,and the rate of bud induction was 86.67%,and the rate of seedling formation was 80.00%,respectively. The most suitable medium for proliferation rooting of Ipomoea batatas variety Xinxiang was MS+0.10 mg·L-1 6-BA+0.05 mg·L-1 NAA. Following the cultivation condition,the number of leaves was 9.50,the number of primary roots was 7.17,the height of the plant was 6.68 cm,the length of root was 38.94 cm,and the stem thickness was 2.46 mm,respectively. We established a comprehensive tissue culture system for stem tip meristematic tissue of Ipomoea batatas variety Xinxiang,laying a foundation for the subsequent development for apical meristem virus-free of Ipomoea batatas variety Xinxiang.
