Study on aseptic culture and adventitious bud induction of two varieties of Jiashi melons

Abstract:

In order to protect the variety and innovate the germplasm of the Jiashi melon(Cucumis melo L.), this study obtained aseptic seedlings through aseptic sowing of common and hybrid Jiashi melon seeds, and explored the conditions for inducing callus tissue and adventitious bud differentiation from different explants. The results showed that the optimal disinfection method for two kinds of Jiashi melon seeds was to soak them in 70% ethanol for 30 seconds and 3% NaClO solution for 3 minutes after shelling them on a clean bench. The best inducing explant was cotyledon of the 5-day aseptic seedling near the hypocotyl. The best induction medium was MS+1.0 mg·L-1 6-BA, with a callus tissue induction rate of 100.0%. The adventitious bud differentiation rate of common Jiashi melon was 73.3%, and that of hybrid Jiashi melon was 66.7%.
