Tolerance and culture condition optimization of hypoglycemic Lactobacillus rhamnosus

Abstract:

This study compared the hypoglycemic abilities of three strains of Lactobacillus rhamnosus and selected Lactobacillus rhamnosus JYLR-005,which demonstrated the best hypoglycemic function,for further investigation. The tolerance characteristics of Lactobacillus rhamnosus JYLR-005 were evaluated through acid tolerance,bile salt tolerance,and survival rates in simulated intestinal fluid and simulated gastric fluid. Additionally,the culture conditions for the strain were optimized. The results indicated that Lactobacillus rhamnosus JYLR-005 could grow in media with pH values ranging from 1.5 to 6.5,showing a higher survival rate at pH value 6.5,which increased over time. The strain could survive under ox bile salt concentrations of 3-9 mg·L?1,indicating a certain level of bile salt tolerance. The survival rates of Lactobacillus rhamnosus JYLR-005 in simulated gastric fluid(pH value 1.5)and simulated intestinal fluid(pH value 6.8)gradually decreased over time. Orthogonal experiments revealed that the optimal culture conditions for Lactobacillus rhamnosus JYLR-005 were pH value 7.5,temperature 40 °C,inoculum size 5%,and incubation time 54 h.
