Abstract:

Root segment propagation of Salvia miltiorrhiza Bge. can shorten the seedling raising time, and an appropriate length of root segments can reduce the consumption of seed roots and cut the production cost of seedlings. For this purpose, different planting lengths of S. miltiorrhiza root segments were set in this experiment, namely 5.5 cm,4.0 cm and 2.5 cm(denoted as T1,T2 and T3, respectively),and an equal-volume mixed solution of IBA and NAA under different concentrations were used for germination acceleration,to investigate the effects of 3 root segment lengths on the agronomic traits, yield, and contents of pharmacologically active ingredients of Salvia miltiorrhiza. The results showed that a 100 mg·L-1 mixed solution of IBA and NAA in equal proportions was optimal for inducing germination of S. miltiorrhiza root segments, with a germination rate reaching 94.44%. The main inflorescence length, number of aboveground branches, and leaf area below the main inflorescence of T1 were significantly(p<0.05)higher than those of T3. However,no significant differences were observed among the three treatments in terms of the number of main inflorescence layers, plant height, number of pinnately compound leaves, main stem diameter, fresh root weight and dry root weight. The root-shoot ratio of T2 was significantly greater than that of T1 and T3. No significant differences were found among the 3 treatments in terms of plant harvest rate, yield per plot, and yield per 667 m2,and the yield of T1 reached 1 204.40 kg per 667 m2. The contents of tanshinones and salvianolic acid B in all treatments exceeded the requirements stipulated in Pharmacopoeia of the People's Republic of China 2020. These findings indicated that planting length of root segments has no significant effect on the root yield of S. miltiorrhiza, and the contents of active ingredients meet the requirements. Therefore, the length of root segments can be reduced when planting, combined with pre-germination induction using growth hormones.
