Abstract:

Continuous cropping obstacle is one of the common problems during continuous cultivation of Pinellia ternata, which causes the yield and quality of Pinellia ternata to decline, and seriously hinders the healthy development of Pinellia ternata industry. Isolation and identification of plant growth-promoting rhizobacteria (PGPR) from the rhizosphere soil of healthy Pinellia ternata grown under continuous cropping were carried out in this study, and their seed germination-promoting effects and growth characterization were also validated. The isolation of the strain was carried out using the plate method. PGPR was screened out using Shanghaiqing seeds and Pinellia ternata seeds, and the growth-promoting characteristics were identified and evaluated based on 16S rDNA sequence analysis, phylogenetic analysis, and morphological feature analysis. The results showed that a total of 109 strains were screened from Pinellia ternata, among which BXGJ053 strain had the effect of promoting the germination of Shanghaiqing seeds and Pinellia ternata seeds, and had a significant(p<0.05))effec on the germination rate, root length and plant height of Pinellia ternata. The results of 16S rDNA sequence analysis and phylogenetic analysis of BXGJ053 strain showed that the strain clustered with Priestia megaterium, and it was named as Priestia megaterium BXGJ053. BXGJ053 strain had the ability to lyse phosphorus, produce ion carriers and IAA. This study provides a theoretical basis for the ecological cultivation of Pinellia ternata.
