Abstract:

To investigate the changes in the composition and structure of soil bacteria, fungi, and archaea under different planting durations in the genuine producing area of Hangbaishao, and to screen for beneficial microbial strains in the rhizosphere of Hangbaishao in this area, rhizosphere soil samples of Hangbaishao aged 1,2,3,and 4 years were collected from Dongyang City,Zhejiang Province. Microbial community structure analysis was conducted to elucidate the microbial composition characteristics and succession patterns in the rhizosphere soil during Hangbaishao colonization. Functional predictions of the microbial communities were performed using the EggNOG,KEGG,and CAZy databases. The results showed that the microbial composition of Hangbaishao soil was dominated by bacteria(85.561%-91.680%),archaea(0.481%-2.804%),fungi(0.018%-0.030%),and viruses(0.022%-0.117%),with other microorganisms accounting for 7.516%-11.500%. As planting duration increased, some inferior microbial groups gradually disappeared, and microbial diversity decreased. The relative abundance of bacteria increased from 85.561% in the 1-year-old rhizosphere soil to 91.680% in the 4-year-old soil. The relative abundances of archaea and viruses declined from 2.804% and 0.117%(1-year-old)to 0.481% and 0.022%(4-year-old),respectively. The relative abundance of fungi first increased and then decreased, from 0.018%(1-year-old)to a peak of 0.030%(3-year-old),and then dropped to 0.024%(4-year-old). Furthermore,CandidatusRokubacteria and Nitrospira were identified as signature taxa in the rhizosphere soil in the genuine producing area. These results indicate that with increasing planting duration in the genuine producing area of Hangbaishao, microbial diversity decreases, while characteristic taxa such as CandidatusRokubacteria and Nitrospira are recruited and enriched.
