Effects of different gibberellin concentrations and stratification temperatures on seed germination of Polygonatum sibiricum

摘要:

本研究探讨了不同赤霉素浓度及温度层积处理对黄精种子萌发的影响,旨在为黄精的规模化、工厂化育苗提供技术方案。结果表明,100 mg·L-1赤霉素处理能够有效打破种子休眠,发芽率高达61.75%。此外,赤霉素与低温沙藏相结合处理时的效果最佳,在15 ℃、100 mg·L-1赤霉素条件下,种子的发芽率达到最高值。在沙藏56 d后,将种子转入泥炭土中培养10 d,其发芽率达到86.51%。此外,随着沙藏时间的延长,种子的萌发时间明显缩短,且种子内脱落酸(ABA)浓度呈下降趋势,也间接表明其萌发率与ABA含量呈负相关。该研究为黄精种子的保存和萌发提供了优化条件和技术支持。

Abstract:

This study explored the effects of different gibberellin treatments and stratification temperatures on seed germination of Polygonatum sibiricum, aiming to provide a technical solution for the large-scale and factory-based seedling planting of P. sibiricum. The results showed that treatment with 100 mg·L-1 gibberellin could effectively broke seed dormancy, and the germination rate was as high as 61.75%. In addition, the treatment effect is the best when gibberellin is combined with low-temperature sand storage. Under the conditions of 15 ℃ and 100 mg·L-1 gibberellin, the germination rate of seeds reached the maximum value. After 56 days of sand storage, the seeds were transferred to peat soil for cultivation for 10 days, and the germination rate reached 86.51%. Furthermore, with the extension of sand storage time, the germination time of seeds was significantly shortened, and the concentration of abscisic acid (ABA) in seeds showed a downward trend, which also indirectly indicated that the germination rate was negatively correlated with the ABA content. This study provides optimized conditions and technical support for the preservation and germination of Polygonatum sibiricum seeds.
